What I would do is close every last program, and then try shutting it down. If it still is resistant, go into your Task Manager, and make sure the Applications tab is completely clear of applications. If any are left, click on End Task for them.
Try shutting down again - if it still won't shutdown, go back into Task Manager and click on the Processes tab. Click on the CPU column here, until the processes using the most CPU appear at the top. If you see any processes using a lot of CPU, end those processes.
If you've tried all this, then do what others have suggested, and hold the power button down on your computer for about 10-15 seconds. If that doesn't work, try shutting the computer off with the button the power strip, if you have one.
Warning: Do NOT pull the power cord out of the wall when your computer is on. You will most likely damage your computer if you do this.

Hold down your power button to overide the cumputer itself. When you restart your computer i would advise doing a system defragment. Also be sure not to open more programs than your system can handle. You will know you computer is working to hard if it starts to slow down while your working,.

I believe that if you hold in your power button for about 10 seconds or so, your computer will automatically shut off.

You can try a forced shut down by typing
Shutdown -f
This should close all applications without warning.
If that does not work, the last resort is to pull the plug :)
