There is really not a best way. Once you submit your story on Oprah.com you just have to hope that it is interesting and/or inspiring enough for her to contact you or even have you on her show.

Through her producers at her email address that is listed on www.oprah.com. She will read the really goods ones, but her staff do all the reading of mail and emails.
