You must get a law degree, pass the bar exam for whichever state you live in and then apply to the Public Defenders office.

In my county, all of the recently licensed (recently admitted to practice) lawyers are given most of the court-appointed criminal defense cases. They act in instances wehre there are conflicts [such as when there are multiple defendants to a crime and one of the defendants is already represented by the Public Defender's Office] and where there aren't sufficient people in the Public Defender's Office to handle cases.

Whenever there is a big drug bust in the county, a lot of lawyers get called up to do court-appointed criminal defense work.

Uh, get a 4 year degree, then take the LSAT, then apply to law school, get into law school, complete law school, take the Bar Exam, apply to a local city or county DA office....

Go to law school, pass a bar exam, and get hired as a public defender.
