It may contain small numbers of white cells, epithelial cells, if you're male, there may be a few sperm in there (depending on the last time you ejaculated). Centrifuging the urine generally concentrates all this into a small pellet, yielding enough DNA to analyse.

This depends on what you are testing. For example, if you are testing the function of the kidneys, you going to be looking for protein/amino acids in the urine. This is often due to a malfunction in the distal tubule. If you are drug testing, your going to look for remnants that have been detoxified by the liver. Another example, is that alcohol is converted to ethanol by the body and therefore is present in urine.
