Well I've gotta confess, I'm a pretty mellow person to start with, so it doesn't take much to make me content, calm and fulfilled.
That said, it's several things. I try not to get stuck in a rut. I seek out new experiences. I try never to lose my sense of curiosity. I try to learn at least one new thing about life every day. (The kind of stuff they don't teach you in school, too!) I'm also happily married, and I'd probably not be as happy if I were still single. My wife contributes a lot to my life's happiness. :-)
The great thing about one's purpose in life is that your purpose is yours to create. Finding it may not be easy, but that's why we have free will and individuality.
Try taking up a hobby, something you may not have considered before. Go for a hike at a forest preserve. Volunteer for some kind of charity. Find something that interests you (not necessarily an academic subject), and learn as much as you can about it. Help a friend or acquaintance through a tough time, and don't be afraid to lean on someone else when you need to. And don't sweat the small stuff.

A hobby will give you all these things.
I learned when I was very young, my hobby was going on vacation! Then, I went with my parents to the mountains of North Carolina and Tennessee. We would drive constantly, stop only for gift shops, dinner and hotel, RARELY stopped at any road-side attraction, but I loved every minute.
Now as an adult, my vacations are limited to 1-3 days. We go to places like Orlando, Tampa, Ft. Lauderdale, Miami, the Florida Keys.
I do not care how hot, cold, wet, dry, crowded it is, I am contented, peaceful, and calm when on vacation.
When I was first married (still married, 30 years), my wife and I would take 2-week vacations. But after a while, that was just too long. We slowly narrowed them down to 1-3 days. Just enough to enjoy ourselves, but not too long to make us crazy!!
30 years ago we drove from West Palm Beach to Key West, 5 hours one way! in a MG sports car, and $35 in cash. Left at 4:30 AM, arrived 9 AM. Drove home at 5 PM, got home at 10 PM. Exhausted, but WHAT a day!! We did not even have a camera!! But STILL remember that day.

The best way to feel better about yourself and your life is help someone else. It's especially rewarding if you can see the joy you might bring, but even in secret, it helps. It REALLY helps if you do something very special for someone who really doesn't deserve it.
If that doesn't work, take a long hot shower in the dark with a candle and lavendar salts on the side of the tub to relax and feel peace.

Listening to the rain on my window as I lay in bed at night. The coo of the morning dove on waking.
The rainbow after an afternoon shower. Walking in the sand barefoot. A childs face when they are discovering something new. A babys smile. A hug.
Quiet time. Time with family and friends. .and it goes on and on.

My trust in God, my 7-year old son and my husband. These 3 make me happy. God has blessed me with happiness and contentment. I pray the same for you. Your life does have purpose, you have to figure it out and do it. If you are good with kids, work with them, if you are good with the elderly, visit a nursing home. You have no idea what you will mean to someone.

Adventure : playing like a child but while mountain biking, or trecking...I did a 4 day treck in the Swiiss alps alone once...ended up spending 2 days in a hut with blindness from the glacier...!
For me, its the reason to be alive. Adventure.
All the rest comes with it. Beauty, calmness, fullfill., success, gratitude....

i find peace, contentment and calmness in the belief that we are here for a reason. i know that that whatever life brings, it all happens for a reason -- even if i don't know yet, what that reason may. try reading the four agreements -- i really enjoyed the book. and yes, i believe in god.

The key is to realize your dreams in life.
I, for one, have set myself to the simple goals of building a modest career and enriching my relationship with God, and my family and friends. Seeing yourself fulfilling these goals gradually is quite contenting. It rid me of the sense of emptiness you have forementioned. Most importantly, it gave me a firm purpose in life to stand on.
