If you are going to Disney Land Orlando
If you are going to Magic Kingdom - make sure you go on space mountain. It is the most fun ride in the park but I think there is a height limit. If your kids are too small, make sure to take them on the swirling tea cups ride. Splash mountain is also fun and traveling to the island where Tom Sawyer lives is also a blast. There is also a great rollercoaster in toontown (based on Goofy). The Pirates ride is also really cool.
If you are going to Epcot - make sure you take them on the information ride through the giant ball. It is a very interesting experience. ALso be sure to take them around the world (Around the lake) so that they can experience different cultures. Other than that, the Finding Nemo aquarium, the thrilling racetrack coaster, and the Bill Nye show were my favorite attractions.
If you are going to MGM - make sure you ride the Tower of Terror and see that fantastic show at the end of the day. There is only one time Fantasmic! plays so you must make sure you watch it. THe courtyard for the show is huge so there will be more than enough seats. Also be sure to go on Star Tours and the classic movie ride. Plus, a round of Who wants to be a Millionaire (you may win something) is fun.
If you are going to Animal Kingdom, make sure you go on the Kali River Rapids and the Dinasour rides. They are both really good. Other than that, Animal Kindom boast a large number of animal attractions (hence the name). You do not want to miss the African Safari, the Pangani Jungle trail and the Maharaja jungle trial, or any other amazing attractions. Also, the Festival of the Lion King show is extremely fun to watch. The new Everest ride is also pretty sweet and the conservation center and petting zoo are great for kids.
If you are going to California adventures, you do not want to miss the shows at Hollywood Backlot.
I do not know much about Disneyland Paris and Tokyo but I bet they are enjoyable experiences at those places as well.
Make sure to get Fastpass tickets early in the morining. Lines at disneyland are deadly!
The hotels, although lavish, are not worth the ridiculous prices. Stay at most one night.
One last note: All of the parks are a blast so do not worry about not having fun!

A few things, well first, be happy, don't let anything ruin your trip. Second, go to the Epcot ball, and there is a ride inside of it. See the parades, go on the Tower Of Terror ( my favorite!), The Pirates of the Caribbean, all of those rides, go to Epcot and see every world, and get food at every place, not some ting big, but something from that country,. Just go to everything you can! But those are like the important ones. (PS. See the parades either on the first day your there or the last day, and go catch the short lines during the parade!) Have fun!

I am a disney World fanatic so it is hard to pick just one thing. I would have to say be sure not to miss the Fantasmic show at MGM which is shown near closing time. Either get there early for a seat or go to guest relations first thing in the morning to get tickets for the Fantasmic dinner package. With this dinner package, you eat at one of the restaurants then get special seating tickets for the show. Have a great time. With DisneyWorld you can't go wrong!

Your question leaves too much open. How old are you? Kids? Ages? Are you a thrill seeker? Or more of an observer? How much time do you have?
I think that the Disney employee has answered best so far.
But...these are a few of my favorite things:
The Lion King Show is spectacular
Everest is a great adrenaline rush.
Epcot is a great way to view the colors and traditions of the world.
And the Animal Kingdom "Nah-ta-zoo" is pretty amazing!

The fireworks are a great ending to a long day. I went last week, and to be honest with you, I was pretty unimpressed!
The fireworks, and the Christmas store in Liberty Square is very nice. If you have never been on Pirates of the Caribbean, it is also a highlight (I didn't ride it this time, but I was told they put Johnny Depp all over it - not sure how I feel about that!).

If you are a coaster fan, Everest at Animal Kingdom, if you are a show person, The Lion King at Animal Kingdom. If you like to walk around exhibits, World Showcase at EPCOT. If you want food, any of the countries in EPCOT. My personal favorite is the France Bakery. Best photo op, Magic Kingdom in front of the castle. So it just depends on what your personal amusement park style is.

It is the entire disney experience and the memories you have after that. Don't miss taking pictures with your most favorite disney character. Don't miss the parade during the day and the fireworks by the castle at night.

Epcot - The night show that brings out all the disney characters on a boat ride with fireworks, singing,dancing etc.
The most memorable to say the least.

Wow.. there's sooo much to do in Disney World, it's ridiculous. You can't even stay for a week and do and see everything in the park.
Most parks close at about 10-11... but at Disney World there is ALWAYS something to do. You're pretty much pampered if you get good packages and stay in a resort. I loved it... I stayed for a week and my parents made sure we had a good stay.
Just try not to miss anything, really. haha.

best ride = animal kingdom safari
Dont stay at disney hotels it's not worth the money
park at downtown disney and ride bus to where you are going ie free parking
