While the most obvious answer being in our time would be Mariano Rivera, let's go back a decade or two and say Dennis Eckersley. Already a dominant starter then converted to one of the best closers that ever lived.
Notables:
Mariano Rivera - yeah ok fine give it to Mr. Roboto
Trevor Hoffman - best changeup EVER!! used to be paired up with a 98MPH fastball, thats just lethal
Lee Smith - he did hold the saves record for a while
Eric Gagne - 82 straight saves without one blown? not bad...not bad at all
Rollie Fingers - the man was a master

Mariano Rivera hands down. He did it during the season but made his money in October. He turned Yankee games into 8 inning affairs for over a decade. If it wasn't for John Wetteland blocking from the closers role early on he could easily be the all time saves leader. As much as I dislike the Yankees, Mo is the clear choice for closers.

I would have to say Mo Rivera of the Yankees.
He's curretly 4th all time in save with 416, won a world series mvp in 1999 ( Only the second closer to do so. The other was Wetteland who's job he took ). And most of all he has unreal post season numbers.

If i was selfish, i would say Bobby Thigpen (1990 57 Saves Folks) but im only kidding.
Rivera was good
Hoffman Is Good
But surprisingly i would want Billy Wagner as my closer. I just love how the guy has electric stuff and a cocky attitude.
Question...Didn't Billy Wagner use to be a righty but he messed up his arm and started throwing left handed? I heard that someplace before. Dont know if its true or not it was waay before he made it to the Show.

Trevor Hoffman holds the record for saves, so I'd have to say him. With 492 saves and still pitching, this guy is headed straight to Cooperstown and is the best closer of all time.

The main 3 are Goose Gossage, Dennis Eckersley and Mariano Rivera. Papalbon will be good also lets not forget Trevor Hoffman or Billy wagner. Both are excellent in their own right

As a Met fan I hate the Yankees but I have to give credit where credit is due. Mariano Rivera is hands down baseball's all time best closer.

Rivera this year. But at one time it was Lidge of the Astros until his meltdown after the NLCS a couple of years ago. It would be difficult to say of all times because batters are getting better.
of course I remember (showing my age) a time when the starter usually went the whole game.
