Masturbating will not deplete your body of testosterone. Spermatogenesis and production of testosterone, which is a hormone that has effects all over the body, are separate issues.

So, no, you don't "waste" any by masturbating. You do lose your sex drive temporarily after you ejaculate, but that's just because you burn up all of your testosterone and endorphins at once when you have an orgasm and your testicles kind of go into spasm and stop producing any more testosterone for a little while (called a refractory period).
