Politically ,Winston Churchill was he most critical opponent and he was forced to resign after the fall of France and the defeat of the BEF in 1940. Churchill was asked to become Prime Minister by the existing War Cabinet.

Chamberlain was forced to resign as Prime Minister when war broke out in Europe after Hitler invaded Poland. After all, Chamberlain had tried to peacefully "appease" Hitler in order to avoid conflict.
So, his biggest political rival was Winston Churchill, who had been telling everyone the whole time that Hitler was going to start a war.
