Some of the ladies here at work have heard that diamonds need a hole underneath them so they can breath, some said they heard that if they don't they are fake. Who knows why they have a hole underneath them.

It's so the light can pass through, so the gem's facets sparkle. No, diamonds do not need to "breathe." Nor are totally enclosed diamonds necessarily fake.

If they have a Di-cast mounting the bottom will have a hole in it. that means if the mount is one solid piece and not a Head (holding the diamond) and a shank (the band) Older rings are typically that way. I was told by an old manager that the hole was an air hole for the diamond. Newer mountings have gaps under the stone.
The best way to test your diamond is to take it to a jewelry store and have them put it on a Diamond Tester.

The setting has a hole under the diamond so you can see through it and enjoy the gem's clarity & faceting.

Only for cleaning!!! Diamonds do not reflect light from underneath Light enters the top and reflects back through the top.

yes they are real is they have a hole to breathe, they can not be enclosed
