Block Island is a quieter place. Nantucket and Martha's Vineyard are equally beautiful but offer more in the way of restaurants, bars, shopping.
Where are you coming from? If you are flying into Boston, I'd suggest taking the ferry to Provincetown. You won't have to rent a car and thus you will avoid the traffic.
It's a great place for both gays and straights with lots of beaches, restaurants, bars and shopping.
Go to a good travel site like skyauctions.com, expedia.com, priceline.com or hotel.com and start looking now.
I could give you recommendations but places book quickly on both the lower and upper Cape as well as the Islands.

Hi - Nantucket is kind of like the Vineyard except that it has this sort of wistful, edge of the known world kind of feel - like you are being watched all the time by something ancient. Makes you want to swim to the bottom of the harbor and see what the pilings are planted in. And the seagulls sing in a different key there. Their eyes are a little wilder. I would tell you about the pine trees, what they do in the fog, but you probably wouldn't believe me. The place is full of Ents.
Nantucket really is a great place to visit in the Summer - or in the early Fall. Crowd disappears right after Labor day, but a lot of the places close down too, so if you want a slow vacation, go after Labor Day. Don't take a car, expensive, and you don't need a car there. Rent a bike, walk, take a bus or hitch hike. From here on, this is a repost from an answer I gave to someone else:
If you are an outdoor person, some good places to see are:
1)Cotue - rent a boat and take a lunch - head across the harbor and have a lot of beach wilderness all to yourselves.
2) Take a drive through the moors - miles of dirt roads through heath & bayberry Approximately 1/3 of America's heath lands (moors) are on the Island of Nantucket.
3) Go out and take a walk in the State Forest or the Hidden Forest.
4) Windsurfing is great in the harbor. Out by Pocomo is a good entrance.
5)Walk the town and the wharves - lots of shops and art studios, nice yachts.
6)Definitely go out to Siasconset - so many amazing homes to see - do some walking around out there.
7) If you are into museums - there is the whaling museum in town, the Nantucket Life Saving Museum out on the Polpis road
8) There are great bike paths - see enclosed links
9) Golfing
10) Hiking - either along the beach, or just drive until you see something interesting - Out towards Wauwinet is good. Beware of poison ivy! Also keep in mind that Nantucket has a BAD tick disease problem - and tics are very active around Memorial Day. Lyme disease, babisiosis, and a few others.. Nasty! Take precautions. The offending tics can be very small - deer tics.
Nantucket is riddled with restaurants and clubs. The Ships Inn has a very good restaurant.. also a nice place to stay. Although i have never spent a single night in a hotel on Nantucket, so I am not the one to answer that part of the question. But the enclosed links should help.

Nantucket is wonderful...I love it there and miss not going as often as I use to when I was living in Boston. But I go back once a year.
If you have an artist nature you will love it here...everywhere you look there are painters. I also love the slow way of life and how pretty the flowers are there...I can always relax on Nantucket Island.
