Thats a virus, get your self an anti-virus program like avg

The files on your pen drive are probably permanent files stored there by the manufacturer for a reason. Or, the drive just doesn't have exactly 40GB of data. Its usually just rounded there. For instance, my jump drive has 3.8GB of data but is is advertised as 4GB of data.

yes it is a virus. Mostly trojan... or which spreads everywhere...

there would be another file 'Autorun' delete it . But by now it is on your computer, so better get an antivirus and get rid of it.

I don't think it's an easy one to remove....
