we do this one often - it's not a bad drive (although, you should avoid rush hour like the PLAGUE)
You're about 75ish minutes to the homestead area (this all depends on traffic through hollywood and miami) and from there it's about three hours.
I actually prefer this option (we usually fly into Miami, though) to flying directly into EYW - depending on how long you're in Key West, the drive through the other keys is just as enjoyable - stop for a drink at islamorada (the tiki bar will soon become high-rise condos, and it's a legend in it's own time), see if you can spot some keydeer on big pine, enjoy a fish sandwich at one of the excellent restaurants along the way and enjoy the scenary.
