You can, you need to have that song hosted on a server... just search Google for "mp3 hosting" when you finish uploading the song they will give you a link to it. You can send that link on a Myspace Comment.

Its a little confusing to do, but you can do it through flash. Either by making a lil flash using the song, and then uploading that flash file to the Internet, and using special html code, it will show up as a little media player on comments.
You can also do this using Windows Media player, or similar embed player.
Can be time consuming.
