The largest transport of all was the ocean liner "Queen Elizabeth" of about 84,000 tons. At the start of the war she was converted to a troopship, and regularly crossed the Atlantic carrying 15,000 or 16,000 servicemen. She and the Queen Mary, her slightly smaller sister ship, travelled without convoy or escort, relying on their sheer speed and pale grey camouflage paintwork to evade the German U-boats. They were called the "Grey Ghosts".

I'm not sure just what you are asking, so I will try to give you several answers.

Literally the largest transportation vehicles were the two Cunard liners, Queen Mary and Queen Elizabeth, which--at 30,000 tons plus--could carry a considerable load across the Atlantic. In both the Atlantic and Pacific most military goods and supplies moved by sea, making sea transport the main overseas carrier in use during World War II. Not that much has changed--in a modern conflict sea transport is still the principle carrier across oceans.

The largest aircraft in use during World War II was most likely a seaplane, such as the Boeing 314 Clipper. Howard Hughes Hercules--or Spruce Goose--would have been the largest, but it wasn't finished until after the War.

Over land the main transportation means was either rail or truck, depending on where you were and the general condition of the roads and railroads. In Europe the rail system was quickly repaired and put into use. Most of Asia didn't have adequate railroads for military use.
