Could be Mumbai - Pune. because, all buses going towards HYD/BLR and umpteen tier - 2 cities nearby, Mumbai - Pune tickets are sold in each and every bus.
If you are curious about longer routes, Mumbai - Bangalore would be the the most busy as it is high revenue grossing, highly profitable route. No convenient trains and airfares are way higher compared to overnight buses.
