To me it means that you can fool me or lie to me once and if I fall for the same lie then I am the fool.
It can also mean that if you "fool" me once I will not believe you in any other thing you tell me.

It has two slightly different meanings. One is that it's normal to take someone's word at first. So, if that person lies, shame on them. They are a liar. Once we know that, we don't have to trust them again. We can forgive but then not find ourselves depending on their words and left vulnerable.
A variation on the phrase is trusting in general. We start out life a bit naive and have pet beliefs. Again, we are lied to and feel bad about it, but it's not our fault. Once we have "lost our innocence", however, we must not keep trusting and believing the same things. We develop habits and wonder why we can't move on with life. We pick the same poor choices over and over. Shame on us.

Well I don't disagree...but in the real life that doesn't count...sometimes you just have to let others fool you...I mean you know they fool you...let them until they want to harm you...and then take action and do something about it.

This expression shifts responsibility for action from where it ought to be.
If you fool me twice, you are likely being dishonest twice. That is your choice and your behaviour. I won't accept responsibility for someone else's behaviour.
Someone who is prepared to fool me twice is someone I would be better off without.

it means that if u fall for a trick , u shouldnt blame ur self because its ur first time and u live to learn , but if u fall for the same trick again , then its ur fault and u should blame ur self for not learning ur lesson the last time :)
