The Small Business Administration has a state by state listing on how to get a business license. Go to the SBA site and select your state http://www.sba.gov/hotlist/license.html

As for the DBA, go to your local city/county clerk's office and apply for a DBA. Some counties have their procedures for applying for DBA online, even the process can be done online. So check if your county has their DBA procedure on their county website. Here are examples

Travis County TX http://www.co.travis.tx.us/county_clerk/dba.asp
Harris County TX http://www.cclerk.hctx.net/coolice/default.asp?Category=AssumeNames&Service=an_inquiry
Monroe County NY http://www.monroecounty.gov/clerk-dba.php
Clinton County MI http://www.clinton-county.org/clerk/dba_search.asp

Ignore the advice of the second person who said that for DBA you have to form a corporation. You can be a sole proprietor and need an assumed name. DBA is merely getting permission to use a name other than your given name (e.g. instead of Jane Doe you name your company as Jane's Apple Growing Company)

For a business license you have to apply to the local village where you want to do business. For a DBA you have to form a corporation and register it with the state and the IRS and then apply for a DBA and pay the fees too.
