James Dean was a good actor. But I'm not sure that you could actually 'label' any one person the greatest of them all. Especially when you consider all of the character actors down through the years and the rich elements they brought to film. I'm not so sure how James Dean would have done with comedy.
I think what James Dean did was bring a new 'style' to acting that hadn't been done before. More subtle. more realistic. It was sort of what the Beatles did for rock and roll. For it's time it was revolutionary and gutsy. This is just my opinion. I'm no expert, just a person who enjoys good movies.
I think Kevin Kline is a superb actor. Even Robin Williams is very good. I am always intrigued with Tommy Lee Jones. He brings a lot of energy to the screen with him. Brad Pitt is really very good also. I have never understood all the hoopla about DeNiro. Meryl Streep was fantastic in Sophie's Choice along w/ K. Klein. And I'll watch anything with Holly Hunter. Robt. Duvall, Anthony Hopkins, George Clooney...they're all good.

You become a lot better than you were if you die young. He was good, very good. But the best ever? No one is good enough to be considered that. I believe in levels. There are plenty of actors who are as good as you can get at the craft.
Gary Oldman, Jack Nicholson, twenty other guys I could rattle off then twenty more I neglected. He didn't live long enough to make a bunch of embarassing movies for the money. He was also a grade A loon and lived in an era where lots of things were covered up. If he had lived I'm sure he would have done some more good work. But he would never have been the legend he is if he died at 84 instead of 24.

No. Not enough roles, no proof he wasn't playing to his limits. Today, based on work range, Johnny Depp and Brad Pitt are probably better (without voting either the best.)
