my friend liz and i have been best friends 4 years and i found out she was lesbian but she does not know i know and i heard her tallking with my boyfriends brother that she was going to ask me out

If you truly are best friends then you should be able to sit down with her and let her know that you know she's a lesbian. She needs to know that she has your support and your friendship. Don't even bring up that fact that she likes you. It may only embarrass her. If she brings it up, just very nicely tell her that you aren't interested in her that way.

if she really is your best friend she will understand if you say `no` and tell her that you dont mind if she likes girls and all that....if you realy dont mind!
