no because the penis is still attached to the body so although the penis is filling up with blood from the body the weight of the penis will still keep the same weight on the scales. now if you are actually wondering if the weight of the penis will be different then the answer is yes because you have the weight of the extra blood to consider but it wouldn't be that much different.

No. The blood that creates the erection just shifts from a different part of the body.
