The way the question is phrased makes me think you should take the quotient first, then do the subtraction. That is, take 100 and divide by 8, getting 12.5, now subtract x and set that equal to 25. If they wanted you to do the subtraction first they might have said "The quotient of one hundred and the difference between eight and a number x is twenty five." Here it indicates that the difference should be part of the quotient.
