The $20 trick does work.
You need to take into account your day of arrival, if it's a Friday or Saturday you're going to have very little chance of a "suite" upgrade.
You might get a room with a strip view >Ask for a room in the Pyramid on an upper floor that faces North< IMHO this is a nice upgrade and worth the $20.
If your arrival is on a Sunday, Monday, Tuesday or Wednesday and does not involve a weekend stay you have at least a chance of an upgrade to a Spa Suite. The Pyramid Spa Suites are some of my favorite rooms in all of LV. Almost 800sq ft, nice Jacuzzi tub built up into the corner of the room, awesome view, very romantic. Since this upgrade "usually" would have a value of at least $100 per night, I would "offer" $50 if this was the upgrade I was asking for.
How to go about "asking"
Have the $$ in your left hand as you hand your ID and CC to the desk clerk with your right. Ask if a "complimentary" upgrade to "whatever you're interested in" is available. He/she will tell you >No/I can't do that/Yes for X amount, in which case you can pony up the difference or forget it. Or, he/she will say, let me take a look for you, in which case you're probably in luck. When he/she hands you the room key to your upgrade, then and only then do you tell him/her thank you and give them the $$ handshake.
It helps to have some knowledge of the hotel you're staying at so that you at least know what to ask for in the way of an upgrade.
The obvious, >be nice, let them do you a favor<

I work the front desk at a hotel....This probably won't work. Most of the rooms are the same for the price you pay. And msot of the time the hotels have it planned out ahead of time what room you're going to stay in. The best thing to do to get a better room is complain about something in your room. lol. I know it sounds funny, but I upgrade people for it. Besides...it doesn't cost me anything. Luxor is a pretty nice hotel though, hopefully you'll just be happy with what you get. (and by the way, I know people say to stay in the tower, but stay in the pyramid. you're right off the casino floor, and the slanty windows are cool).

I used to work at the Luxor.....they will ask you if you want to upgrade because they get a commission of the upgrade price. Giving the front desk person a little "thank you" doesn't hurt either. How much you want to give is up to you. You want a room in the towers though. They are nicer then in the pyramid, especially if it should rain when you are in town. The rooms in the pyramid tend to leak due to the angles.

I've never seen that work. Most room are assigned by a computer. Try this instead: call the hotel directly and ask for the name of the front desk manager and the fax number. Write a letter to the manager and ask if there's anything they can do to make the trip more memorable or more special - translation: upgrade. (Tell them it's your first time to Vegas or a special event or it's your first time at the hotel.) The very least you'll get is a free meal.

Just stay in the Tower rather than the Pyramid and you'll be all good. More room in the rooms (no slanted window) and quieter and less smokey.
