By car it is about 25 minutes depending on the route you decide to take.
I-4 is the easiest and usually the quickest but if there is an accident or back-up for any reason it could take closer to 45 minutes if not more.
International Drive wouldn't be bad if you don't mind the constant stop go traffic due to people not knowing exactly where they are/want to go and the traffic lights more or less designed to help those stop to bring business into the surrounding businesses.
Here are two other routes if you really just want to deal with the local traffic and will be a lot quicker then I-Drive and sometimes I-4.
1) From Walt Disney World Property
*Take Buena Vista Dr (the road infront of Downtown Disney) all the way to SR-535 (away from parks), you'll see Crossroads, take a Left at the light here
*At the next light take a Right onto Palm Parkway (eventually turns into Turkey Lake Rd)
*Stay on this road until you reach Universal Studios on you're Right hand side

2)From Walt Disney World
*Take a Left onto SR-535
*Once you reach Sand Lake Rd take a Right
*Take a Left onto Turkey Lake Rd
*Universal Studios will be on your Right

Everyone is correct in saying it's approximately 20 minutes by car, providing the traffic isn't backed up. But just to let you know, the majority of the hotels in the area offer free shuttle buses to all of the Disney parks and to Universal Studios... even SeaWorld, if you are interested in that as well :)
