I took this same trip from Houston, Texas (at legal speed limits) here is the information below:

Driving distance: 377.7 miles
Trip duration: 6 hours, 23 minutes
Driving time: 6 hours, 23 minutes
Cost Approximately: $44.07
Sorry, I was driving a Mitsubishi Gallant 4 cyclinder sedan
and with rising gas prices now it should be an extra 10-15 dollars added to above price depending on grade of gasoline you may be using, also keep in mind this info is for one way.

From Houston to san antonio it's about 3 hours, from san antonio to south padre is another 3. So 6 hours? but there may be a quicker route. mapquest it.
