I'm not sure what the first thing they did was, but one of the early things they did was to rip all the beds out of their ships and start using hammocks, like the Native Americans.
I'd imagine that other things high on their list was to
1. give thanks that they found land
2. expore
3. gather whatever they could to take back
4. Claim whatever they found for Spain
5. Grab a few Native Americans to take back with them
6. Document their findings.

Once they saw the land, and thought they had discovered it, they soon found out that there were already natives living there. The rest, as they say, is History.
