Its a new type of transport system to allow more information to travel back and forth between the motherboard's CPU and the PCI-Express Video Card.

Most PCI Express cards are video cards. There are a few other devices that add to the video card Physx cards are 1 such device.
Also Crossfire (ATI) and SLI (nVidia) are 2 PCI-express slots on the same motherboard that link the cards together creating a very high def image.

PCI express slot is for a graphics card which supports 16 X data transfer speed. Before PCI we had AGP which could support a maximum of 8x , also graphics card supporting PCI is cheaper that the same model supporting AGP.
