I've been to all of the places listed above me and I agree they are great. But of the places I've been, Barrow, Alaska really is the most unique for me. I was there the first week in April, the Arctic Ocean was frozen and the wind chill was -58 F.
I have not yet been to Isle Royale NP, but hope to get there someday.

We have visited Neil Armstrong's boyhood home in Wapakoneta, Ohio.
There is a Museum there which has many items on loan from NASA. Even his back-up suit for Apollo 11.
Talking to the volunteers who staff it was more interesting than the displays!
We have also visited the "Field Of Dreams" movie location site in Dyersville, Ohio.
The farmer had previously replanted over the field but so many people kept coming, he had to remake the baseball field.
Famous and popular, but it fills your "unique" requirement to a tee; Mount Rushmore in South Dakota.
Awesome - everyone should visit it. And while there go south a ways and visit Crazy Horse - as yet unfinished.
Caddilac Ranch in Texas, you need to see this for yourself to appreciate how different and amazing it is:

The National Parks are obvious choices
Crater Lake
Glacier
Sequoia
and of course the Grand Canyon are ones I would pick out.
However, Dead Horse point state park, Craters of the moon NM and Lava Beds NM are also awesome places to visit.

The Mammoth Cave (Kentucky)
Did You Know?
Mammoth Cave is the world's longest known cave. In fact it is so long that if the second and third longest caves in the world were joined together, Mammoth Cave would still be the planet's longest cave and have over 100 miles left over!
