what 4-letter combination can be pronounced differently depending on the letters in the rest of the word.

and no i am not referring to the f-word either.

This is not really a four letter word but it is four letters that can vary enormously depending on which word they are in.
O-U-G-H
Cough - OFF
Dough - OH
Bough - OW
Thought - OR

One such word could be TEAR.
To indicate a cut, it is pronounced as teh(r) or as 'tare'.
To indicate the salty secretion from the eyes, it would be pronounced as 'tir' or as 'ti(u)r'.
