This depends if you want to stay on the strip or not.
Do you want to stay at a hotel that has a casino?

If you are just looking for a nice place to stay check out any of the major hotel chains web sites, they all have places all over Las Vegas.

Last time i stayed at a Clarion Inn. I was only 1 block from the strip, I had a nice room with a kitchenete, living room and bedroom for only $98.00 a night.

Remember the time of year, and the day of the week will depend on what rate you pay.

If you are going and want a once in a lifetime experience, do spend the extra money and stay at one of the big resorts on the strip.

I did that the first time and it was nice, but since I'm not a big gambler, having a casino in the hotel is not a big deal to me. Also I always rent a car so I can drive to anyplace I want.

AS FAR AS A THE BEST HOTEL:

The Orleans
was rated "Best Value in the US" ( by tripadvisor.com users)
rated "Best Bargin 2006" (by Las Vegas Journal)
ranked #2 for "Best Hotel" in Vegas (by Yahoo users)

its clean
it has spacious rooms
14 great restaurants
great gambling
friendly staff
nice pool & spa
bowling alley
movie theater
less than a mile from the Strip
free shuttle every 15 minutes

If you must stay on the strip...
I'd say all of the major Casino are nice.

Pick a price you want then pick a theme you like and would enjoy.

The Best Hotel and the best price seldom go together.

When booking you should consider the most important things
.
1. location to be near what you hope to do.
2. Facilities like restaurant, pool and tour desk in each hotel if you want to do more than sleep there.
3. Additional services like airport and around Vegas shuttles.
Hope you enjoy your trip.
Cheers!

Sahara is definitely a good choice if on a budget. Can get a room under $100 for the weekend (see story below). Regarding best hotels, their prices go way up, probably as expensive as you want ....

I think the Imperial Palace is the best value and location. A balcony room in the middle of the strip. Have stayed there a dozen times

Emerald suites Tropicana, it is a little off the strip but there is a free shuttle that will pick you up and drop you off... they have a pool, jucuzzi, and their rooms are set up like an appt. they have a living room, full kitchen and bedroom, i booked them through priceline for 75 a night on athurs and friday which is usually more expensive

if you are in the military, or have friends at Nellis AFB, you can stay there at $30.00 dollars a night, but if you dont, i would stay at alladin, or circus circus. The best is probably Mandalay Bay, but it is very expensive there.
