Engine misfire.

Vacuum leak?
Look around the intake manifold for a small vacuum line that is not hooked up, or a vacuum port without a hose hooked up to it, or capped off and is sucking air.

Loose spark plug wire?

Improperly gapped spark plug?

With the engine cold, start it up and let it idle for about one minute.
Now, with a bottle of windex or other non-flamable sprayer, spray the exhaust manifold at the engine and which ever exhaust port does NOT sizzle, that's the one that's misfireing.
Pull that spark plug and see if it's gapped to .036"

ADDED:

I seriously doubt it's transmission related.
If the engine is running, the tourqe converter is spinning whether it's in gear, or park.

It could be a lot of different things ,a bad torque converter is one but I don't get why it doesn't do it when its in park.Make sure you don't have a vacuum leak.Block all the hoses coming of the carb, see if that helps.Double check your firing order, timing, also make sure your engine mounts and trans mounts are good and also tight.Good luck to you.

i own a repair shop ,and first you need to find out if its a vibration or a miss-fire in the engine,i think it may be out of balance and be shaking, and if that is the case with it you,ll have to pull the transmission back out of it and change the torque converter in it, that's the only way to balance it back out,but find out for sure if its an actual miss-fire or just a vibration ,that would help a lot in answering this one, good luck with it hope this helps.

I would suggest checking all vacum lines
and make sure it has the correct gasket under carb
