Hard to pick one, because each successive project brought knowledge and technology that didn't exist before that project.
The most documented and "historical" event would have to be the moon landing in 1969. But there have been some amazing missions since then, most notably (in my opinion) the Mars Rovers - they have been there for almost 3 Earth years, more than twice the initial estimate.

Well, putting a man on the moon was definitely up there. There's also NASA's 'big four' observatories to consider - Hubble, Chandra, Compton Gamma Ray Observatory, and Spitzer. Oh, and putting multiple probes on Mars. That was pretty cool.

Apollo of course. Also Gemini and Mercury. Also Viking and Voyager. And the Space Shuttle of course. The space station is international, but the U.S. is the largest contributor.
