It's a tossup between the giant squid in Disney's "20,000 Leagues Under the Sea" (1954), the great and awesome beast created by the Disney techs that attacked the submarine "Nautilus" in the middle of a raging storm, and the absolutely terrrifying and disgusting plague of monster sea-snails that came roiling up from the sea in the black-and-white 1957 production, "The Monster That Challenged the World".
Much as I've enjoyed Ray Harryhausen's creations of sea-going horror (the Kraken, the five-tentacled monster octopus from "It Came From Beneath the Sea"), I still have to give my vote for the first two I've mentioned.
