You'll need to be a little more specific to receive a good answer.
If you're talking about web browsing, you can look in the History area of the web browser. If it's empty, you know that no one has browsed the Internet from that program, or they've went to the trouble to delete the history when they are done.
Now, if you're talking about people using various programs on your computer, thats not quite so easy. You may find some activity logs in the Administrators Tools that are helpful, but sometimes not.
There are programs that you can add onto your computer that secretly record activity and you can look at it remotely or when you return to your PC.

Probably the easiest way is is to check for new files or recently modified files. You should go to search for a file then choose option when it was modified (put the dates you are interested in) and you will get every file opened / modified on that date. There is also other option: right click on my computer and choose manage option. When new window opens just check for events in event viewer. Have a look down there and you will know almost everything.

Go to Start - control panel - user accounts. Make sure your account is Administrator and set a password - and don't give it to anyone. Delete the Guest account and set up a limited user account. Let your mates use that, they can't see your files unless you put them in the shared folder.
If they set a password to lock you out you can log on as Admin and fix it or even delete the account.

Download a key logger that hides in the background, that way you can tell what has been viewed and also if they have used any of your user names and passwords to look at certain sites. If they have you know that you need to change your passwords and/or lock your Pc when you are out.

open the internet and click on History and if there are sites that you've not been to there then you'll know.
if you don't want anyone on there at all make a password for your computer.
open start, go to control panel, user accounts, click on you account, make password.

if u feel someone is using the computer when they should not,
put a power on password (access through the bios) and no one is going on your system...
otherwise use the key logging software... because if they get into your stuff, why not get into theirs(fair game)... hopefully they are using your system to get into their emails...

Would you believe it, just look into history, this is one area they forget to delete.
If you are talking about your files, etc, then go to search, search for files that have been accessed during the period you have been away and away you go.

you should have Mcaffee Privacy Service set up, that way someone else cannot access the internet on your computer without typing in a code word.
