I must add to Molly's statement.
When God asked Cain: "Where is Abel?", Cain replied, "Am I my brother's keeper?"
So, Cain was asking, in defiance, 'Am I in charge of protecting my brother?' although Cain and God both knew the truth: that Cain had just killed his brother Abel.

It refers to the answer Cain gave to God in Genesis when God asked, "Where is Abel?" Of course, He knew Cain had murdered his brother. In writing, the allusion means someone responsible for another.
