The most famous play is undoubtedly Hamlet. It's been translated into nearly every language (including Klingon).
As for my favorite... I've got a soft spot for Much Ado About Nothing. It's not his best play, but it's the only comedy that's actually funny just from the text. (Midsummer is also hilarious, but mostly from the physical comedy that you add around the text, not the text itself.) Other plays are more powerful, but Much Ado is the most fun.

I feel it has to be, 'Romeo and Juliet'. It's a house hold word and many who do know he wrote have no clue what else he wrote.
Hamlet would probably be the most famous amongst academics and theater goers.
Personally I like 'Taming of the Shrew'. From the standpoint of choreography I like the Tempest.

Romeo and Juliet
The name Romeo has even become part of the language to mean a hopeless romantic, and the story itself has been retold and revisited hundres of times, much more than any of his other works.
My personal favorite is Richard III...such a great villian and the work is a great morality play.

Midsummer's Night Dream is the best. My favorite lines are:
Love looks not with the eyes, but with the mind. And therefore is winged cupid painted blind.
Lord, what fools these mortals be.
For this their jampering I esteem a sport.
Their navery is to make an *** of me

Hamlet. Queen Gertrude is an awesome part. She goes from being innocent to her son going insane and then telling her that her current husband killed her first husband, then getting killed herself.
Plus it's easier to understand and not boring like Julius Caesar.

My favourite is "Twelfth Night." I love all the depth there is to all the characters, and the plot is very good.
Although I am generally opposed to modern revisions of Shakespeare plays, the "Twelfth Night" with Parminder Nagra was just delightful. I especially loved how seamlessly they used an Anglo/Indian cast.
