I mean when they are eating in the fancy restaurant in the hotel, vesper is leaves because she looks on her phone and then she says "mathus needs me"
Then a few seconds later, james says "mathus???" and knows something is wrong,
What caused him to figure out that she is in danger?
Like what did he think, and what caused him to think this? I missed out on that detail

He realized Mathus worked for Le Chiffre, because Le Chiffre found out James knew his "tell" (when he was gambling and bluffing, he would move his hand toward his eye). The only person who could tell Le Chiffre that James knew his tell was Mathus, because he was the only one James told besides Vesper.
Bascially Bond realized Mathus was working with Le Chiffre, when Le Chiffre changed his "tell" inexplicably.

And just in case the audience didn't get that...when he was pulled out of the wreckage, the bad guy said...
"It turns out that your friend Mathis was actually MY friend Mathis."
