I have heard several speculative explanations, as to what is actually in the briefcase, but there is one which seems to stand out in my mind, as being the more interesting poignant.
You recall the band-aid on the back of Marsailles Wallace's neck??? Many biblical references reportedly dictate that when the devil takes your soul, he removes it from the back of your head. Now, if you recall, the combination of the briefcase is '666', and when anyone opens up the case, a brilliant, overwhelming light cascaded from it.
Is it possible that inside the case was Wallace's soul, and perhaps he was trying to buy it back??? The three kids in the apartment, who had the case (including ol' 'Flock 'o' Seagulls') were the devil's couriers; when one of the kids flew out of the bathroom, and fire a 'hand cannon' at Jules and Vincent, all of the bullets 'missed' them. "God came down and stopped these M/F bullets", because they were here to save Wallace's soul, hence the 'devine intervention' cognigtation.
An interesting speculation, to say the least, but as I said before, most poignant, would you say???

Yeah, Tarantino purposefully left it ambiguous as he felt a reveal would be dissappointing. Popular thoughts are that it is Elvis' Gold Jumpsuit, which played a role in Tarantino-written True Romance, the diamonds from Reservoir Dogs, just generic gold, and, most popularly, Marcelas Wallis' soul (hence the bandage on theback of his neck and the fact that 666 is the combination). Ultimately though, the case is just a plot device.

the case is a red herring. it is used to distract you from the film. some say that it contains the soul of marsellaus wallace hence the band-aid on his neck. some say it may be the gold elvis suit that was stolen in true romance, or the glowing contents of the malibu in repo man.

Its up for you to decide. In a radio interview with Howard Stern in late 2003, Quentin Tarantino was asked by a caller the contents of the briefcase, and he answered, "It's whatever the viewer wants it to be." which is so gay I know I want a really answer!!!!!!!!!!!!!!!!!!!!!!!

no one knows. Tarantino deliberately left the contents of the case secret so that there would be this discussion about it.

There's deliberately never been an answer to that question. We're supposed to guess what it is.
