You need to know some, probally for the fact that no one would take you seriously otherwise. You do need an incredible knowlege of astrophysics and a body that could handle the stress of multiply G's. It is also a good guess to say that you have to know how to fly a plane, since almost all of them are military pilots.

Maybe not the whole universe but some knowledge of where you were going to travel to would be useful.

Not really. Most astronauts are former military pilots.
