Alan Shepard was the first American (human) into space, aboard Freedom 7 on My 5th, 1961. The trip lasted approx 15 minutes.
However, prior to that in the early days of space exploration, nobody knew if people could survive a trip to space. Using animals was the best way to find out. In 1948, a monkey named Albert flew inside a rocket. Nine years later, the Soviet Union sent a dog named Laika into orbit.
So actually, the first American into space was an animal.
