Four
Sedin Twins
Staals (Yes Eric & Jordan are brothers, with Marc could be playing more next year.)
Niedermayers
Hossas
WOW! There are currently a lot! Looks like more.
Fedor Federov doesn't play in NHL, neither do more than 1 Messier as far as I can see.

I've got 14 sets. Here they are:
1. Pascal & Phillipe Dupuis
2. Scott & Rob Neidermeyer
3. Henrik & Daniel Sedin
4. Mikko & Saku Koivu
5. Marcel & Marian Hossa
6. Frantisek & Tomas Kaberle
7. Milan & Zbynek Michalek
8. Roman & Vojtech Polak
9. Aaron & Ashton Rome
10. Eric, Jordan & Marc Staal
11. Paul & Yan Stasny
12.Mark, Mike & Colin Stuart
13. Chris & Josh Gratton, and
14. Jussi & Kimmo Timonen.
They are all listed as being assigned an NHL team even if they did not play in the NHL this year. Hope that is a good answer.

There are 11 sets of brothers currently playing in the NHL;
Scott and Rob Niedermayer of the Anaheim Ducks, Henrik and Daniel Sedin of the Vancouver Canucks, Robin and Richie Regehr of the Calgary Flames, Tomas Kaberle of the Toronto Maple Leafs and Frantisek Kaberle of the Carolina Hurricanes, Marian Hossa of the Atlanta Thrashers and Marcel Hossa of the New York Rangers, Saku Koivu of the Montreal Canadiens and Mikko Koivu of the Minnesota Wild, Tuomo Ruutu of the Chicago Blackhawks and Jarkko Ruutu of the Pittsburgh Penguins, Henrik Lundqvist of the New York Rangers and Joel Lundqvist of the Dallas Stars, Eric Staal of the Carolina Hurricanes and Jordan Staal of the Pittsburgh Penguins, Milan Michalek of the San Jose Sharks and Zbynek Michalek of the Phoenix Coyotes, and Paul Stastny of the Colorado Avalanche and Yan Stastny of the St. Louis Blues.
Neither Mathieu Biron or Kevin Smyth (if he is Ryan's brother) played in an NHL game this season, so they would not count. Neither do Staffan Kronvall and Niclas Kronvall, as Staffan did not play a game for the Maple Leafs this season.

I can think of three sets of brothers in the NHL: the Staals, the Sedins, and the Birons. There are probably more, though.

Don't forget that Peter Schaeffer of Ottawa's brother is Nolan Schaeffer, the goalie on the Penguins minor league team that they acquired at the deadline from the Sharks. I don't believe Nolan played at all this year but he played well last year.
