It is hard to tell if a turtle is gravid. sometimes you can see bulges at the lower end of the turtle just above the rear feet. You can have the x-rayed for error proof testing. The 55 gallon tank should have 1/3 substrate and the rest water. Check out this link. http://www.tortoisetrust.org/articles/Nestsites.htm
Hope this helps. Good luck!

I'll assume you know you have a female turtle, and that it is old enough to be sexually mature- about 4" long.

The most reliable indicator of eggs is an X-ray, but if the eggs are well-developed we can feel and sometimes even see the bulges around the rear legs.

For tank set-up, try http://www.austinsturtlepage.com/Care/caresheet-eastern_painted.htm for some ideas. It is tough to do this in an indoor tank, but you can set up a plastic tub with a mix of clean soil, sand, and mulch that the female can get into and out of.

The Painted Turtle is a common North American turtle. The adult turtle ranges in length from 4-10 inches. The carapace is a smooth, flattened oval, and is green to black in color. The plastron is generally yellow, sometimes tinged with red, sometimes with a black to reddish brown figure of varying size and shape. The skin of the Painted Turtle is black to olive with red and yellow stripes on the neck, legs and tail and yellow stripes on the head. Males have elongated foreclaws and long, thick tails. Females have shorter foreclaws, shorter and thinner tails, and tend to be larger.

Turtles lay eggs, so you need some dry sand above the waterline.

Have never figured a way to tell if one is about to lay eggs though. The shell doesn't swell up.
