Three important factors that limit cell size are:
1. Surface area to volume ratio. As cells get larger, the volume increases at a faster rate than the surface area increases. The problem is that there isn't enough surface area or membrane to handle exchange of molecules for a big cell.
2. Rate of diffusion is too slow to move materials to the interior of a large cell as the materials are needed.
3. DNA cannot be copied (transcribed) fast enough to give instructions to make enough proteins for a big cell.
