The First World War (1914-1918).

This particular war either directly or indirectly caused the Russian Revolution of 1917, the rise of Hitler and the Nazis in Germany, the Second World War, the nearly total destruction of pre-1914 European civilization and the Cold War between USSR and USA.

It was the war that made the 20th century the hellhole that it was.

Assassination of Franz Ferdinand. This led directly to WW1, and WW1 changed everything. 4 empires disappeared, 2 of which had been around since the Middle Ages. WW1 caused the Bolshevik Revolution in Russia, which led to the establishment of the world's first Communist state. Europe lost its economic, cultural, and moral position as the world's leading continent. WW1 was the beginning of the world we know today, so the assassination of Franz Ferdinand was the most significant event of the 20th century - the other 2 were effects of WW1.

For the world to lose that many people, some of whom may have been the ones to cure the common cold, or to be great leaders, was cataclysmic.
