Something that is bespoke is something that is unique and specially made purely for the purpose it serves. It is not copied from anywhere nor is it used for any other purpose other than the one it was specifically made for.

Just like a bespoke painting is made just for the person who comissioned it, bespoke web designs are unique and will never be used again for another site.

There are two types of application.
Bespoked and Off-the-shelf.

Bespoked is something that is designed and written for a specific need. It does exactly what you want.
Off-the-shelf is an already written application. It may not fit the required need but the department or company will have to change to make the application work for them.

So a bespoked development is one that is specially created for the company. It's like does not exist anywhere else.
