That was the presumed time that Ronald Defeo jr killed his family and that is why "George"kept waking up at that time. In the movie Ronnie Defeo was supposed to be driven mad by the evil in the house and he started killing his family at 3:15 am. So the movie wants to give the same impression that the evil is trying to drive George crazy and coax him into repeating the same crimes at the same exact time.

3:15 is also considered by science to be the time inwhich most 'nightmares' take place in the human brain. It's a known fact that more people wake up in the middle of the night at 3:15 than at any other time. The nightmare explaination is the only one ever given for this. I, myself, always wake up at 3:33am in the middle of the night and the only connection I can tell is that I was born at 3:33pm in the afternoon so maybe it's my own body's 12 hour clock waking me up. But, anytime I do wake up at 3:33, I'm always coming off a really vivid dream. ;-)
Now, non-scientist believe 3:15 to be the 'bewitching hour'. Which is probally why they use it in the movie.

One more fact to add is that 3 AM is considered the start of the "witching hour" by Demonologist everywhere including Ed & Lorraine Warren, the real life investigators that were involved in the haunting. The witching hour, 3 AM, is when demonic activity in a haunted house typically begins.

George would wake up around 3:15 every morning and would go out to check the boathouse. Later he would learn that this was the estimated time of the DeFeo killings.
