I've Seen This Question A Hundred Times And Once Again No They Are Story Line Half Brothers But In Real Life They Are Not Related At All Although They Are Close Freinds. Undertaker Is Mark Calaway Kane Is Glen Jacobs.

Kane is a character portrayed by Glen Jacobs. Glen himself is in no way related to Mark Calaway (the man who portrays the Undertaker.) As the story goes, Kane is Taker's half brother, Taker's mother had an affair with Paul Bearer and Kane was the result of that.
It was first said that Taker torched his family's funeral parlour which killed his parents and his brother Kane. Paul took Taker under his wing until 1996 when he turned his back on Taker, telling him he had "dirt" on him.
As it turns out, Kane didn't die in the fire, he was scarred, but lived. He had watched his brother rise to fame in the WWF (now WWE) and wanted to be in that spotlight too and eventually face his brother and destroy him.
When Kane was brought to the WWF, he was a masked character, supposedly to hide the burn scars on his face.
After a few years of feuding, Kane was unmasked by Taker and it was obvious that Kane did not have such facial scarring as first believed. It came out that the scars were emotional, not physical.
Over the years, Kane and Taker have taken on each other in numerous matches and even formed a tag team, Brothers of Destruction.
All of this has evolved Kane into the character that we currently know. Again, the character played by Glen Jacobs.
