Here's what you need to do.
1st, don't waste your time with AVG, spybot, ad aware, CA, spyhunter, superantispyware or anything of the sort. Stay away from anything that is FREE when it comes to spyware and antivirus. There is a reason why they free. They are NOT good.
2nd, You need some of the best software out there. Norton Systemworks is very good. That's the only good software from Norton. I wouldn't recommend Norton Internet security, its not that great. Now the best software you should get is Spysweeper with Anti-virus, this software is too good and gets better everyday. This is elite software. It blocks over 173,000 threats. No other software can compete with that or come close for that matter. It catches everything including Spylock. I've used them all and it is 2nd to none.
3rd, after you install and update the software, since its in your system you will have to turn off system restore and then go into safe mode and run the scan and eliminate the virus. then after removal, reboot and turn system restore back on.
4th, (Optional) I recommend a Firewall from Zone Alarm, their free version is so good. Or you can buy their Pro version, but the free version will do more than the job.
5th, any probs during or after you do all this, send me an e-mail.

You have one of the latest variants of the Smitfraud infection. Here is a link to complete removal instructions.

How to remove SpyLocked (Removal instructions using the free Smitfraud Fix removal tool):
http://www.bleepingcomputer.com/forums/topic85376.html

Most "general-purpose" anti-malware programs such as Norton, AVG, AdAware, etc. can not or will not remove this pest Smitfraud. And most who would tell you such, have no actual factual information. They are simply touting there favorite anti-virus or anti-spyware as as "cure for everything".

BTW, SpyLocker usually gets installed when someone goes to view a video at a "not-so well-known" website. The website will say that a "special" codec is needed to view it, and asks to download it to you. If you allow the download, you will NOT be downloading a codec, you will actually download a trojan called Zlob. The moral is, never allow a website to download "stuff" unless it's a well known and respected site. And oh yeah, that video would have played just fine without that "special" codec!

Good luck!

SpyLocked is the latest rogue anti-spyware program to come out from the makers of SpyDawn and Virusburst. SpyLocked is installed by a trojan called Zlob, which attempts to trick you into buying the alleged anti-spyware program. Once you're infected with SpyLocked, a fake security message similar to a Windows notification pops up saying your PC is infected with malware. This SpyLocked message is used to lure you into purchasing, downloading and installing their program to remove the imaginary spyware.

SpyLocked may also automatically launch at your computer's startup and scan your computer. SpyLocked can be extremely difficult to remove manually, and will continue to try to recreate itself.

SpyLocked is a Dangerous fake antispyware software and it is update version of SpyDawn. SpyLocked is a variant of Trojan.Zlob. SpyLocked display a fake warning message to purchase the paid version of SpyLocked.

SpyLocked also displays a fake warning alert with flashing icon on your system tray. A Pop up balloon warning messages claiming that your PC is infected. For example : "Critical System Error", "Your computer is infected", "System Alert", "Security Alert", Trojan-Spy.win32@mx", "Virus Alert", "Security Alert" or "Spyware.Cyberlog-X".

I got all these information from this site and you can also check it. There you can also get the complete manual and Automatic removal.
