the most turns the game will give you is 1000. At that point, it evaluates the score, culture, etc. (whatever limits you have set) and then awards the victory.
When you pick your nationality at the beginning, click on game limits to make some of these changes. Frankly, playing for the trip to Alpha Centauri gives you a more clear cut victory. So likely will some other options like capture the princess, but I haven't tried the latter.
The other thing is that when it says the game is over, it will also ask if you wish to continue play. No more points, but if you're trying to militarily defeat everyone, you can keep going.

Yes, you can turn it off. That said, you are misinterpreting it. If you are unable to "win" by the 2050, then you've failed. The time-frame is part of the conditions for winning. If you turn it off, then it just means you were unable to beat it within the context of the winning conditions. It is like extending a baseball game to extra innings just because you are losing.

Civ 3 is a turn-based game, not real-time. The year progression is part of the game. The end year is determined by your choice of length of a game. You can't turn it off, but you can adjust what year your game starts at (5000 B.C., 4000 B.C., 3000 B.C.) When it hits that end-year, the winner will be determined by score.
