Well, you're well on your way. The BIOS supporting it is the most important thing.

So, you have an .iso file. These are called "image" files. When they are in this format, they contain a compressed version of whatever is in the file, in your case they contain a compressed version of the operating system. You will need to unzip the .iso file onto the pen drive so, when booting, the system can read it.
To do this, you will need a program that can extract .iso files. I like WinRAR (http://www.winrar.com), which can open many other files, ISOBuster (http://www.isobuster.com), or Undisker (http://www.undisker.com). All these programs are free to use, WinRAR being shareware with a 40-day use period, after which it will nag you to buy it.
Download one of the above programs, and make sure that it is "associated" with .iso files. Now the .iso will have the icon of that program, and when you double-click on it the program will open, sort of winzip-style. Click the "extract" button (or equivalent), and select your pen drive as the destinatino. The program will now extract to the pen drive, provided there is enough space. (Note: Space can be a problem. If you're trying out Linux or some other OS on the pen drive, you can often find a smaller version that will fit, or buy a bigger pen drive).
Once the .iso is extracted, you can now boot from the pen drive. I'm assuming you know how to do that (go into the Setup menu of the BIOS and select the BIOS to look for bootable USBs before hard drives).
Good luck!
